What did the leader of the others mean when he said that the reason why they were running tests on sayid was because there is a "darkness" growing inside him just like what had happened to claire? have we seen or heard of the darkness before in previous seasons?

No, we haven't heard of it before, but it seems to have something to do with the Monster. Claire was last seen with her Dad, who is supposed to be dead, the Monster takes the form of dead people and hates Jacob. They were in "Jacob's" cabin, and Ileana said someone else was using it.

Rousseau said her team was "sick" which happened after they went down the hole where the Monster was, it may have warped their minds as well, but I'm not entirely convinced Sayid and Claire are the "dark" ones.

The Others have a history of lying, and Sayid seems normal. Sure, Claire did kill two Others, and seems to have set those traps in the jungle, but she was probably alone for 3 years, and she killed to save Jin who was/is her friend. If she was really so unstable wouldn't she have just shot him too?

Not exactly. It seems to be similar to what Rousseau described happening to her team. She called it the Sickness. And Desmond and Inman thought they could become infected if they left the Hatch, and the Hatch mural says I AM SICK on it, And don't forget the Quarrantine sign on the inside door of the Hatch.

I think they are referring to the same thing that's happened with Smokey taking over Locke's appearance, but I could be wrong. Could also be related to the Christian that we saw in the cabin and at the Frozen Donkey Wheel in his plaid shirt (instead of the suit that Jack always see him in).

It will be interesting to see if all of these situations have a similar cause. We just don't know yet.

ETA: I don't think Ben is infected, because Ben was revived when the water was clear. Something was clearly wrong with the spa/spring when they put Sayid into it. It was murky and it didn't heal Dogen's hand.

Remember the French Lady? In the first season, she talked about killing her entire team because they were sick. In the fifth season, the flashbacks showed her team, and they were obviously sick. Not physically sick, but they seemed to have turned evil. Also in the fifth season, Sayid shot Ben, and Kate brought Ben to the others. They said that if they fix him "he will never be the same", implying that this "darkness" is in Ben as well. As usual, we will not totally understand this "darkness" until the end of the show, but I imagine it is like being possessed.

I am sorry to say the user right below me is incorrect, as we never saw the water Ben was put into. The episode entitled "Whatever Happened, Happened" ended with Richard taking Ben into the temple. However, we never acutally saw what happened to Ben inside the temple. The next episode, entitled "Dead is Dead", starts off with Ben inside a tent, and he has already been healed. Therefore, we never saw what the water was like. However, if it were clear, I imagine Richard would not have said "his innocence will be lost" and "he will never be the same".
